产品描述:

与原核细胞、酵母细胞以及昆虫细胞相比,中国仓鼠卵巢细胞(CHO)作为宿主细胞表达的外源蛋白最接近其天然构象,因而CHO细胞表达系统是生物工程制药最为理想的表达系统。在CHO细胞中表达治疗性蛋白对于商业化的药物生产十分经济方便,但是这类产品的生产以及纯化过程有可能会带来CHO宿主细胞蛋白(CHO HCP)残留的污染,这些残留的蛋白污染会降低药物治疗效果,造成副作用或者免疫反应,因此要对CHO HCP的残留量进行控制。《中华人民共和国药典》2020年版三部建议HCP采用酶联免疫吸附测定(ELISA)方法检测。本产品是一种双抗夹心ELISA试剂盒,所使用的抗体来自兔和羊的混合抗体,抗原为CHO细胞的发酵上清收集液(细胞培养基不含蛋白),本产品用于多种CHO宿主细胞表达产物的纯化工艺开发、过程控制、成品放行等。
首次使用本试剂盒前建议先完成产品的适用性研究,确认样本基质是否存在干扰以及合适的样本检测稀释条件。

The ResiQuant® Recombinant Protein A ELISA Kit is a highly sensitive and quantitative ELISA kit. This kit is specifically intended for the detection of process-related impurities recombinant Protein A residuals in the manufacturing of therapeutic monoclonal antibodies or other Fc-fusion recombinant proteins. The limit of detection (LOD) for this kit is 13.7 pg/mL, and the limit of quantitation (LLOQ) is 41.2 pg/mL.
